Biological Background: CEP44 Function and Localization
- Centrosomal protein of 44 kDa (CEP44), also known as HBV PreS1-transactivated protein 3 (PS1TP3), is encoded by the CEP44 gene (KIAA1712). This protein exists in multiple isoforms due to alternative splicing.
- Acts as a centriole-enriched microtubule-binding protein essential for centriole biogenesis. Collaborates with CEP295 and POC1B to drive centriole-to-centrosome conversion by ensuring proper centriole wall formation. Related references: PMID:32060285
- Functions as a linker component maintaining centrosome cohesion. Associates with CROCC and regulates its stability and centrosomal localization. Related references: PMID:31974111
- Subcellularly localizes to the centrosome, centriole, spindle pole, and midbody, consistent with its roles in cell division and centrosome dynamics.
- Classified under keywords including 3D-structure, coiled coil, phosphoprotein, and reference proteome, indicating structural features and regulatory post-translational modifications.
Experimental Guidance and Technical Tips
- The antibody was raised against a recombinant fusion protein covering amino acids 1–280 of human CEP44 (N-terminal region). For Western blot (WB), a band at ~44 kDa should be observed; validate using positive control lysates from human or mouse cell lines.
- When performing ELISA, consider using the immunogen as a coating antigen or a known positive sample; titration may be required to optimize signal-to-noise ratio.
- Cross-reactivity with human and mouse CEP44 is reported; verify specificity in your experimental system using appropriate controls.
- For centrosome/centriole studies, immunofluorescence may be attempted; however, this application is not validated for this product, so antibody suitability should be empirically tested.
