Biological Background: CENPK Function and Localization

• CENPK, also known as interphase centromere complex protein 37 or AF-5alpha, is a centromere protein component of the CENPA-CAD complex.
• It plays a critical role in kinetochore assembly, mitotic progression, and chromosome segregation by recruiting the CENPA-CAD complex to centromeres.
• Through coordination with KNL1, CENPK facilitates the recruitment of the NDC80 complex to the outer kinetochore, ensuring proper microtubule attachment.
• Subcellularly, CENPK localizes to the nucleus, specifically at centromeres and kinetochores on chromosomes.
• Tissue specificity: Detected in several fetal organs with highest levels in fetal liver; in adults, it is weakly expressed in lung and placenta.
