  • Cenexin1/ODF2, also known as outer dense fiber protein 2 or cenexin, is encoded by the ODF2 gene and exists as multiple isoforms via alternative splicing.
  • It is a major component of sperm tail outer dense fibers (ODFs), filamentous structures that contribute to the passive elastic properties and motility of sperm.
  • As a centrosomal scaffold, Cenexin1/ODF2 localizes to the distal/subdistal appendages of mother centrioles and is required for the recruitment of PLK1 and ninein (NIN) to the centrosome.
  • The protein is essential for the assembly and/or maintenance of normal centrin-1 (CETN1) architecture at the centrosome.
  • Subcellularly, Cenexin1/ODF2 is found in the cytoplasm, associated with the cytoskeleton, microtubule organizing center, centrosome, centrioles, spindle poles, and within cilia and flagella.
  • Testis-specific expression is observed at the protein level, with high abundance in the cytoplasm of round spermatids and localization to the midpiece and principal piece of the sperm flagellum.
  • Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation, consistent with its regulatory roles in spermatogenesis and cell cycle.

  • This antibody was raised against a synthetic peptide from the C-terminal region (aa 730-829), which may recognize multiple ODF2 isoforms; use this information when interpreting band patterns in Western blot.
  • For Western blot, a specific signal around 95 kDa is anticipated in human testis or sperm lysates, but examine for possible additional bands due to alternative splicing or modifications.
  • In ELISA applications, coating with the immunogen peptide or recombinant ODF2 protein may be used; titrate the antibody and appropriate secondary detection reagents to optimize signal-to-noise.
