Biological Background: CELF2 Function and Localization
- CELF2 (CUGBP Elav-like family member 2), also known as BRUNOL3, ETR-3, NAPOR, and CUGBP2, is an RNA-binding protein that mediates post-transcriptional regulation.
- It regulates pre-mRNA alternative splicing, mRNA translation, and stability. For example, it activates exon 5 inclusion of TNNT2 in embryonic skeletal muscle and controls actinin exon switching. Related references: PMID:11158314, PMID:11931771, PMID:12649496
- CELF2 participates in apoB mRNA editing by binding to AU-rich sequences upstream of the edited cytidine. Related references: PMID:11577082
- It preferentially binds UG-rich motifs, UGUU sequences, and (CUG)n repeats, targeting a wide range of transcripts. Related references: PMID:11158314, PMID:11931771, PMID:15657417
- Together with CELF1, CELF2 negatively regulates miRNA biogenesis, such as pri-MIR140 processing. Related references: PMID:28431233
- The protein localizes to both the nucleus and cytoplasm.
- Tissue expression is broad, with isoform-specific patterns: isoform 1 in brain and lung, isoform 2 in heart, brain, kidney, and skeletal muscle, among others.
- Keywords from UniProt associate CELF2 with disease variants causing epilepsy and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 422–521 within the C-terminal region of human CELF2. Confirm that this epitope is present in the isoforms relevant to your study.
- For western blot, the predicted molecular weight is 54 kDa. Alternative splicing may generate isoforms of different sizes; include appropriate positive and negative controls.
- This antibody is suitable for ELISA. For quantitative applications, consider performing a pairing study with matched detection antibodies.
- Cross-reactivity is reported for human; reactivity with other species has not been characterized and would need independent validation.
