- CELF1 (CUGBP Elav-like family member 1), also known as CUGBP1, BRUNOL2, or EDEN-BP, is a 52 kDa RNA-binding protein that belongs to the CELF family, characterized by three RNA recognition motifs (RRMs) involved in post-transcriptional gene regulation.
- Functions broadly in pre-mRNA alternative splicing, acting as both activator and repressor; for example, it promotes exon 5 inclusion of TNNT2 (cardiac troponin T) pre-mRNA during heart remodeling and regulates mutually exclusive exons in α-actinin transcripts.
- Regulates mRNA translation and stability, including increasing translation of CEBPB in aging liver and CDKN1A by antagonizing CALR3 repressive effects, and mediates rapid cytoplasmic deadenylation of transcripts containing EDEN elements by recruiting PARN.
- Binds to diverse RNA motifs: (CUG)n triplet repeats in DMPK, Bruno response elements (BREs), AU-rich elements (AREs) in JUN and FOS, and muscle-specific splicing enhancer (MSE) sites in TNNT2.
- Localizes to both the nucleus and cytoplasm, enabling its involvement in nuclear pre-mRNA processing and cytoplasmic mRNA fate determination.
- Plays a role in miRNA biogenesis by binding to primary microRNA pri-MIR140 and, together with CELF2, negatively regulating its processing into mature miRNA. Related references: PMID:28431233
- Post-translationally modified by acetylation and phosphorylation, and subject to ubiquitination (isopeptide bond), which may modulate its activity and stability.
