Biological Background: CEL Function and Localization

  • Bile salt-activated lipase (CEL), also designated BAL, BSSL, carboxyl ester lipase, cholesterol esterase, and sterol esterase, is a secreted serine esterase encoded by the CEL gene (Gene ID: 1056) on chromosome 9.
  • CEL catalyzes the hydrolysis of a broad spectrum of substrates, including cholesteryl esters, phospholipids, lysophospholipids, di- and triacylglycerols, and fatty acid esters of hydroxy fatty acids (FAHFAs), with a preference for FAHFAs bearing the ester bond distal to the carboxylate and unsaturated FAHFA species. This activity is essential for complete digestion of dietary lipids and absorption of fat-soluble vitamins in the intestine. Related references: PMID:10220579, PMID:27509211, PMID:27650499
  • The protein is primarily produced in the pancreas and lactating mammary gland, secreted into pancreatic juice and milk, and has also been detected in eosinophils and duodenal fluid.
  • CEL is a glycoprotein that contains multiple disulfide bonds and an N-terminal signal peptide for secretion. Alternative splicing generates several isoforms, and mutations in the CEL gene are associated with maturity-onset diabetes of the young type 8 (MODY8).
  • Keywords from UniProt include secreted, glycoprotein, serine esterase, lipid degradation, disulfide bond, and alternative splicing, reflecting its functional and structural properties.

Experimental Guidance and Technical Tips

  • This polyclonal antibody was raised against a recombinant immunogen spanning amino acids 350–550 of human CEL (NP_001798.2); it may recognize isoforms and orthologs containing this region, including mouse CEL (reported cross-reactivity).
  • For Western blot, consider using pancreatic or mammary tissue lysates, or CEL-transfected cell lines, as positive controls. The predicted molecular weight is 79 kDa, but glycosylation may cause migration shifts; validate in the relevant sample system.
  • In ELISA, confirm reactivity against the full-length protein or recombinant fragment and titrate the antibody to determine optimal coating and detection conditions.
  • When working with mouse samples, verify binding and optimal dilution, as cross-reactivity has been reported but may differ from human reactivity.
  • Optimize assay conditions and include appropriate negative controls to account for polyclonal background.
