Biological Background: CEL Function and Localization

  • Bile salt-activated lipase (CEL), also known as bile salt-stimulated lipase (BSSL) or carboxyl ester lipase, is a serine esterase that catalyzes the hydrolysis of a wide range of substrates including cholesteryl esters, phospholipids, lysophospholipids, di- and tri-acylglycerols, and fatty acid esters of hydroxy fatty acids (FAHFAs). It preferentially hydrolyzes FAHFAs with the ester bond further away from the carboxylate, and unsaturated FAHFAs are hydrolyzed more quickly than saturated ones. This enzymatic activity is essential for the complete digestion of dietary lipids and their intestinal absorption, as well as the absorption of fat-soluble vitamins.
    Related references: PMID:10220579, PMID:27509211, PMID:27650499
  • CEL is secreted and primarily expressed in the mammary gland and pancreas. It is detected in pancreatic and duodenal juice at the protein level.
    Related references: PMID:21784842
  • Expressed also by eosinophils, contributing to local lipid metabolism at sites of inflammation.
  • CEL is a glycoprotein with multiple repeats and contains disulfide bonds, which are important for its structural stability and enzymatic function.
  • Alternative splicing generates multiple isoforms, and mutations in the CEL gene are associated with maturity-onset diabetes of the young type 8 (MODY8), highlighting its role in metabolic disease.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 350-550 of human CEL, targeting a region that may detect full-length protein and potential isoforms. Validate antibody reactivity in your specific sample system to confirm isoform recognition.
  • For Western blotting, the predicted molecular weight is approximately 79 kDa. Consider using reducing conditions to address potential disulfide bond–related mobility shifts.
  • In ELISA, the recombinant fusion protein used for immunization may serve as a positive control. Optimize coating and detection conditions with purified CEL or relevant biological fluids.
  • Cross-reactivity with mouse and rat CEL is reported, making this antibody suitable for translational studies across species. Confirm reactivity in your animal model tissue or cell lysates.
