Biological Background: CEBPE Function and Localization
- CCAAT/enhancer-binding protein epsilon (CEBPE) is a member of the C/EBP family of transcription factors, named also as CRP1, C/EBP-epsilon, and linked to immunodeficiency 108 (IMD108).
- Acts as a transcriptional activator, regulating gene expression during myeloid differentiation. Related references: PMID:26019275
- Recognizes and binds to two distinct DNA motifs: the CCAAT homology sequence found in many promoters, and the enhanced core homology sequence common to many enhancers.
- Essential for the promyelocyte-to-myelocyte transition in myeloid cell development. Related references: PMID:10359588
- Localizes to the nucleus, consistent with its role as a DNA-binding transcription factor.
- Strongest expression is observed in promyelocyte and late-myeloblast-like cell lines.

Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (amino acids 1–190); thus this polyclonal antibody may detect multiple epitopes on CEBPE. Consider using full-length controls to confirm specificity.
- For Western blot, a band at ~31 kDa is expected; include promyelocyte or myeloblast cell lysates (e.g., HL-60, NB4) as positive samples, as these express CEBPE most abundantly.
- The antibody is suitable for ELISA; validate by coating with recombinant CEBPE protein and optimize coating concentrations empirically.
- Cross-reactivity with mouse is noted; for murine samples, confirm reactivity in the target tissue or cell line and adjust blocking conditions as needed.
