Biological Background: C/EBPB Function and Localization

  • C/EBPB, also designated NF-IL6, TCF5, LAP, and LIP, is a CCAAT/enhancer-binding protein beta transcription factor encoded by the CEBPB gene.
  • It critically regulates the expression of genes involved in immune and inflammatory responses, including binding to regulatory regions of acute-phase and cytokine genes (PubMed:12048245, PubMed:1741402, PubMed:18647749). Related references: PMID:12048245 PMID:1741402 PMID:18647749
  • Essential for adipogenesis: after phosphorylation-mediated activation, it induces CEBPA and PPARG, thereby driving the transcriptional program for adipocyte differentiation (PubMed:20829347). Related references: PMID:20829347
  • Plays additional roles in gluconeogenesis, liver regeneration, hematopoiesis, female reproduction, osteoblast differentiation, and intracellular bacteria killing, with functional diversity arising from interplay with other proteins and regulatory modifications.
  • Localizes to both the nucleus and cytoplasm; its activity is governed by post‑translational modifications, including phosphorylation, acetylation, methylation, and ubiquitination, as well as by isoform‑specific interactions.
  • The LIP isoform acts as a dominant negative regulator through heterodimerization with the LAP isoform, influencing processes such as osteoclastogenesis (PubMed:11741938). Related references: PMID:11741938
  • Expressed at low levels in lung, kidney, and spleen, and is indispensable for macrophage activation and CD4+ T‑cell responses.

Experimental Guidance and Technical Tips

  • For Western blotting, the expected molecular weight is approximately 36 kDa; however, be aware that multiple isoforms and extensive post‑translational modifications may cause additional bands or shifts.
  • For immunohistochemistry (IHC‑P), consider using liver, adipose tissue, or inflamed tissue as positive controls, and validate the signal with a known negative control tissue.
  • For ELISA, due to the polyclonal nature of this antibody, include appropriate controls to confirm specificity and consider potential inter‑batch variation.
