Biological Background: CEACAM6 Function and Localization
- CEACAM6 (CD66c, NCA) is a cell surface glycoprotein belonging to the carcinoembryonic antigen (CEA) family, encoded by the CEACAM6 gene.
- It mediates calcium- and fibronectin-independent homophilic cell adhesion, as well as heterophilic interactions with CEACAM5 and CEACAM8, contributing to intercellular contacts. Related references: PMID:2022629 PMID:16204051 PMID:11590190
- Plays a role in neutrophil adhesion to cytokine-activated endothelial cells, facilitating immune cell trafficking. Related references: PMID:1378450
- Involved in tumor progression, promoting cell migration and adhesion to endothelial cells, which may support metastatic spread. Related references: PMID:16204051 PMID:10910050 PMID:11590190 PMID:1378450
- Localized primarily to the cell membrane and apical cell surface, with a glycosylphosphatidylinositol (GPI) anchor.
- Contains immunoglobulin-like domains and forms disulfide bonds; subject to N-glycosylation.
- Expressed in neutrophils, colon epithelial and goblet cells, and various tumor cell lines. Related references: PMID:1378450 PMID:10436421 PMID:16204051
- Keywords also highlight roles in apoptosis and oncogenesis, with a known 3D structure.
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 200–300 of human CEACAM6, likely targeting an extracellular domain. For Western blotting (WB), consider using reducing conditions and whole-cell lysates from CEACAM6-expressing cell lines (e.g., human colon carcinoma HT-29 or MCF-7). The predicted molecular weight is ~37 kDa, though glycosylation may cause band shifts.
- For immunofluorescence on paraffin-embedded sections (IF-P), validate in appropriate tissue types and optimize antigen retrieval protocols. The antibody may also be suitable for ELISA applications; however, cross-reactivity with related CEACAM family members (e.g., CEACAM5, CEACAM8) should be carefully evaluated in assay development.
- Given the reported cross-reactivity with mouse and rat, this antibody may be useful for studies in these species, but additional validation is recommended.
