Biological Background: CEACAM6 Function and Localization
- CEACAM6 (CD66c), also designated NCA or CEAL, is a cell surface glycoprotein belonging to the carcinoembryonic antigen (CEA) family.
- It mediates calcium- and fibronectin-independent intercellular adhesion, engaging in homophilic and heterophilic interactions with CEACAM5 and CEACAM8 (PubMed:11590190, PubMed:16204051, PubMed:2022629). Related references: PMID:11590190, PMID:16204051, PMID:2022629.
- CEACAM6 promotes neutrophil adhesion to cytokine-activated endothelial cells and binds CEACAM8 on activated neutrophils, contributing to inflammatory processes (PubMed:1378450, PubMed:8776764). Related references: PMID:1378450, PMID:8776764.
- It plays a role in tumor progression by enhancing cell migration and adhesion to endothelial cells, and its expression is elevated in numerous tumor cell lines (PubMed:16204051). Related references: PMID:16204051.
- The protein is anchored to the apical cell membrane via a GPI linkage and features immunoglobulin domains, disulfide bonds, and glycosylation sites.
- CEACAM6 exhibits tissue-specific expression in neutrophils, colon epithelial cells, and goblet cells (PubMed:1378450, PubMed:10436421). Related references: PMID:1378450, PMID:10436421.
Experimental Guidance and Technical Tips
- The immunogen encompasses the extracellular domain (aa 35–320), suggesting the antibody recognizes native and denatured CEACAM6; consider initial use in denaturing Western blots with a ~37 kDa expected band.
- For ELISA, coating with recombinant CEACAM6 protein or cell‑based assays may be suitable; optimize coating concentrations and detection conditions for your specific system.
- This antibody is raised against human CEACAM6; cross-reactivity with rodent orthologs is not expected, but verify if needed with species-appropriate samples.
- Given the GPI-anchored nature of the target, include appropriate lysis buffers or enzymatic release strategies if studying membrane localization.
