Biological Background: CEACAM5 Function and Localization

  • CEACAM5 (carcinoembryonic antigen, CEA, CD66e) is a cell surface glycoprotein involved in cell adhesion, intracellular signaling, and tumor progression. Related references: PMID:10864933, PMID:10910050, PMID:2803308
  • It acts as an oncogene by inducing resistance to anoikis in colorectal carcinoma cells, thereby promoting tumor progression. Related references: PMID:10910050
  • Mediates homophilic and heterophilic cell adhesion with other CEACAM family members, such as CEACAM6, contributing to tissue architecture and cell-cell communication.
  • At the protein level, CEACAM5 is expressed in columnar epithelial and goblet cells of the colon and is highly abundant in adenocarcinomas of the digestive tract and fetal colon. Related references: PMID:10436421
  • Functions as a receptor for E.coli Dr adhesins; bacterial binding triggers dissociation of the CEACAM5 homodimer.

  • For WB: Use denaturing conditions. The mature glycoprotein migrates at ~77 kDa, but glycosylation may cause band shifts. Validate with a positive control such as colon carcinoma cell lines (e.g., HT-29, Caco-2).
  • For ELISA: This polyclonal antibody may be employed as a detection antibody after appropriate titration, using recombinant CEACAM5 protein as a standard and a matched capture antibody.
  • Cross-reactivity has been demonstrated for human; assess other species if relevant to your experimental system.
