Biological Background: CDKN3 Function and Localization
- CDKN3 (Cyclin-dependent kinase inhibitor 3), also known as KAP, CDI1, CIP2, KAP1, is a dual-specificity protein phosphatase that belongs to the protein tyrosine phosphatase family.
- Functions as a cyclin-dependent kinase (CDK) inhibitor, dephosphorylating CDK2 at Thr-160 in a cyclin-dependent manner, thereby regulating cell cycle progression. Related references: PMID:7569954
- Exhibits dual specificity, capable of dephosphorylating substrates containing phosphotyrosine or phosphoserine residues. Related references: PMID:8127873, PMID:8242750
- Localizes to the cytoplasm, particularly the perinuclear region.
- Associated with disease variants and plays a role in cell cycle control.
- Alternative splicing generates multiple isoforms of the protein.
Experimental Guidance and Technical Tips
- The immunogen consists of a recombinant fusion protein spanning the N-terminal 1-69 amino acids of human CDKN3, ensuring high specificity for the target.
- For Western blot (WB), the predicted molecular weight is approximately 24 kDa; validate detection in Human and Mouse lysates, as cross-reactivity is documented.
- For immunohistochemistry (IHC-P), immunofluorescence (IF/ICC), and ELISA, consider optimizing blocking and antibody dilution conditions using appropriate positive and negative controls.
- Non-validated applications or species should be tested empirically with proper controls.
