Biological Background: CDKN2C/p18-INK4C Function and Localization
- CDKN2C, also known as cyclin-dependent kinase 4 inhibitor C, p18-INK4c, or p18-INK6, is a member of the INK4 family of CDK inhibitors.
- The protein functions as a potent inhibitor of CDK6 and, to a lesser extent, CDK4, thereby regulating cell cycle progression.
- By inhibiting CDK4/6, p18-INK4C prevents phosphorylation of the retinoblastoma protein (RB), leading to cell cycle arrest in G1 phase.
- p18-INK4C contains ankyrin repeat motifs, which are essential for its protein–protein interactions and inhibitory function.
- Highest expression is observed in skeletal muscle, with additional presence in pancreas and heart tissues.
- The protein plays a critical role in controlling cell proliferation and is implicated in tumor suppression.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 1–120 of human CDKN2C) corresponds to the N-terminal region, which is expected to be conserved across species, supporting cross-reactivity with mouse and rat.
- For Western blot, a band at approximately 18 kDa (observed molecular weight) should be detected; consider using appropriate positive controls such as skeletal muscle or pancreas lysates.
- In immunofluorescence/ICC, optimize fixation and permeabilization conditions to preserve epitope recognition.
- For ELISA, pair with a matched detection antibody to enhance specificity; validate with recombinant protein or cell lysates.
