Biological Background: CDKN2A/p19ARF (Tumor suppressor ARF) Function and Localization
- CDKN2A encodes two distinct proteins through alternative splicing: p16INK4a and p19ARF (the tumor suppressor ARF). The p19ARF protein is unrelated to p16INK4a in amino acid sequence.
- p19ARF acts as a potent tumor suppressor by inducing cell cycle arrest in G1 and G2 phases. Related references: PMID:8521522 PMID:9393858 PMID:15601844 PMID:17936562
- It binds to MDM2 and sequesters it in the nucleolus, blocking p53 degradation and enhancing p53-dependent transactivation and apoptosis. Related references: PMID:10359817 PMID:9529248
- p19ARF also induces G2 arrest and apoptosis in a p53-independent manner by preventing activation of cyclin B1/CDC2 complexes. Related references: PMID:15361884
- It down-regulates BCL6-induced transcriptional repression by binding to BCL6. Related references: PMID:15567177
- Subcellular localization: mainly found in the nucleolus and nucleoplasm; a short-lived mitochondrial isoform exists.

- Additional functions include sumoylation regulation, interaction with E2F1 and MYC, and a role in autophagy and caspase-independent cell death.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide derived from amino acids 1-100 of mouse p19ARF. The antibody likely recognizes the N-terminal region of the protein.
- For Western blot (WB), test the antibody on mouse cell lines known to express p19ARF. The predicted molecular weight is ~19 kDa, but additional bands may appear due to post-translational modifications or alternative isoforms. A positive control sample expressing p19ARF is recommended.
- For ELISA, the immunizing peptide can serve as a positive control. Validate the assay with known concentrations of the peptide to establish standard curves.
- The antibody cross-reactivity is reported for mouse. If working with other species, verify reactivity experimentally.
