Biological Background: CDKN1C/p57 Kip2 Function and Localization
- CDKN1C encodes the cyclin-dependent kinase inhibitor 1C (p57Kip2), a potent inhibitor of several G1 cyclin/CDK complexes including cyclin E-CDK2, cyclin D2-CDK4, and cyclin A-CDK2, and to a lesser extent the mitotic cyclin B-CDC2.
- It functions as a negative regulator of cell proliferation and may play a role in maintaining the non-proliferative state throughout life.
- The protein is also known as p57Kip2 and belongs to the cyclin-dependent kinase inhibitor family.
- It is predominantly localized in the nucleus.
- Tissue expression is broad: high levels in placenta, lower in liver, and detectable in heart, brain, lung, skeletal muscle, kidney, pancreas, testis, and eye.
- Post-translational modifications including phosphorylation and methylation, along with alternative splicing, contribute to its regulation.
- Mutations are associated with Beckwith-Wiedemann syndrome (BWS) and Wilms tumor, and it is classified as a tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-100); validate antibody specificity with appropriate peptide blocking or knockout controls.
- For Western blot, the predicted molecular weight is ~32 kDa; consider using low molecular weight markers and optimize transfer conditions.
- For ELISA, standard peptide coating or indirect detection protocols may be used; titrate antibody concentration for optimal signal.
- As CDKN1C is a nuclear protein, fractionation or nuclear enrichment may improve detection sensitivity in certain sample types.
- Cross-reactivity with species other than human has not been assessed; check sequence homology before using on non-human samples.
