Biological Background: CDKN1C/p57 Kip2 Function and Localization

  • CDKN1C encodes the cyclin-dependent kinase inhibitor 1C (p57Kip2), a potent inhibitor of several G1 cyclin/CDK complexes including cyclin E-CDK2, cyclin D2-CDK4, and cyclin A-CDK2, and to a lesser extent the mitotic cyclin B-CDC2.
  • It functions as a negative regulator of cell proliferation and may play a role in maintaining the non-proliferative state throughout life.
  • The protein is also known as p57Kip2 and belongs to the cyclin-dependent kinase inhibitor family.
  • It is predominantly localized in the nucleus.
  • Tissue expression is broad: high levels in placenta, lower in liver, and detectable in heart, brain, lung, skeletal muscle, kidney, pancreas, testis, and eye.
  • Post-translational modifications including phosphorylation and methylation, along with alternative splicing, contribute to its regulation.
  • Mutations are associated with Beckwith-Wiedemann syndrome (BWS) and Wilms tumor, and it is classified as a tumor suppressor.
