- CDKN1A/p21CIP1 is a cyclin-dependent kinase inhibitor that plays a critical role in controlling cell cycle progression and DNA damage-induced G2 arrest (PubMed:9106657). Related references: PMID:9106657
- The protein acts as a key mediator of p53/TP53-dependent inhibition of cell proliferation in response to DNA damage, and also participates in p53-independent G2 arrest pathways in certain cell types.
- p21 binds to and inhibits the activity of cyclin-dependent kinases, preventing phosphorylation of critical substrates like RB1 to block cell cycle progression; it also facilitates nuclear localization and assembly of the cyclin D-CDK4 complex, with its effect on kinase activity being stoichiometry-dependent.
