Biological Background: CDKN1A/p21CIP1 Function and Localization
- Cyclin-dependent kinase inhibitor 1 (p21) is a potent inhibitor of cyclin-dependent kinases, crucial for controlling cell cycle progression at the G1/S and G2/M checkpoints. Related references: PMID:9106657
- It mediates p53/TP53-dependent inhibition of cellular proliferation in response to DNA damage, and also participates in p53-independent G2 arrest through CREB3L1 in specific cell types such as astrocytes and osteoblasts.
- p21 binds to and inhibits cyclin D-CDK4, cyclin E-CDK2, and cyclin A-CDK2 complexes, but at low stoichiometry, it can also facilitate the assembly and nuclear import of cyclin D-CDK4 complexes, thereby promoting RB1 phosphorylation.
