Biological Background: CDKN1A/p21CIP1 Function and Localization
- Cyclin-dependent kinase inhibitor 1, designated CDKN1A, is also known as p21, CIP1, WAF1, SDI1, CAP20, CDKN1, and MDA-6. It functions as a potent regulator of the cell cycle.
- CDKN1A plays a critical role in p53/TP53-mediated cell cycle arrest in response to DNA damage, blocking G1/S progression and enabling DNA repair (PubMed:9106657). Related references: PMID:9106657
- The protein binds to and inhibits cyclin-dependent kinase (CDK) complexes, preventing phosphorylation of key substrates such as RB1, thereby halting cell cycle progression.
- At low stoichiometric ratios, CDKN1A promotes the nuclear localization and assembly of the cyclin D-CDK4 complex, enhancing its kinase activity toward RB1, whereas at higher ratios it inhibits the complex.
- CDKN1A competes with POLD3 for binding to proliferating cell nuclear antigen (PCNA), resulting in inhibition of DNA synthesis by DNA polymerase delta (PubMed:11595739). Related references: PMID:11595739
- The protein primarily localizes to the nucleus but is also found in the cytoplasm. Its nuclear localization is essential for CDK inhibition, and its activity is modulated by post-translational modifications including phosphorylation, acetylation, and ubiquitination.

- For immunohistochemistry (IHC-P), both nuclear and cytoplasmic staining may be observed. Optimize antigen retrieval and antibody dilution using known positive control tissues such as colon carcinoma or proliferating cells.
- In ELISA applications, use recombinant p21 protein or cell lysates from p53-activated samples to establish standard curves and validate specificity.
