Biological Background: CDKN1A Function and Localization

  • CDKN1A (p21) is a cyclin-dependent kinase inhibitor from the Cip/Kip family, originally identified as a CDK-interacting protein, melanoma differentiation-associated protein, and a p53-inducible factor.
  • It mediates G1/S and G2 cell cycle arrest by binding to and inhibiting cyclin-dependent kinases (CDK2, CDK4/6), thus blocking RB1 phosphorylation and E2F1-dependent gene transcription (PubMed:36689330). At stoichiometric levels, p21 facilitates cyclin D-CDK4 assembly and nuclear import, while at higher concentrations it inhibits CDK4 kinase activity (PubMed:25329316). Related references: PMID:36689330 PMID:25329316
  • It plays a central role in DNA damage-induced G2 arrest through both p53/TP53-dependent and p53-independent mechanisms, including CREB3L1-mediated arrest in astrocytes and osteoblasts (PubMed:37178686). Related references: PMID:37178686
  • CDKN1A competitively binds PCNA, displacing POLD3 and thereby inhibiting DNA synthesis by DNA polymerase delta.
  • It localizes to both the nucleus and cytoplasm, consistent with its roles in cell cycle regulation and cytoplasmic signaling.
  • Expression has been confirmed at the protein level in keratinocytes.
  • Keywords associated with this protein include acetylation, phosphorylation, ubiquitin conjugation, zinc-finger domains, and metal-binding, reflecting complex post-translational regulation.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this rabbit polyclonal antibody is full-length recombinant mouse CDKN1A (aa 1-159), which is expected to recognize multiple epitopes and both unmodified and post-translationally modified forms of endogenous p21 in mouse and rat samples.
  • For Western blot analysis, an 18 kDa band is predicted; however, due to post-translational modifications (e.g., phosphorylation, ubiquitination), additional higher molecular weight species or shifts may be observed.
  • The antibody is supplied for WB and ELISA applications. In sandwich ELISA, consider pairing this polyclonal as a detection antibody with a validated capture antibody.
  • p21 expression is typically low in cycling cells but strongly upregulated following DNA damage. Consider treating samples with DNA-damaging agents (e.g., etoposide, doxorubicin) or serum starvation to induce p21 before testing.
