Biological Background: CDK8 Function and Localization

  • CDK8 is the catalytic subunit of the Mediator complex, a large coactivator required for transcription of nearly all RNA polymerase II-dependent genes. Mediator bridges gene-specific regulatory proteins and the basal transcription machinery.
  • Acts as a serine/threonine-protein kinase that phosphorylates the C-terminal domain (CTD) of RNA polymerase II, inhibiting transcription initiation complex formation. It also phosphorylates CCNH, leading to TFIIH down-regulation and transcriptional repression. Related references: PMID:10993082
  • Interacts with MAML1 to hyperphosphorylate the NOTCH intracellular domain, promoting its degradation and modulating NOTCH signaling. Related references: PMID:15546612

  • The immunogen corresponds to the N-terminal region (aa 1-100) of human CDK8, which is highly conserved among mammals, supporting the observed cross-reactivity with mouse and rat.
  • For Western blot, a band at approximately 53 kDa is expected. Optimize lysis buffer and sample preparation to account for CDK8’s incorporation into the large Mediator complex.
  • When using ELISA, consider pairing with a detection antibody that recognizes a distinct epitope or a post-translational modification to enhance specificity for the activated kinase.
