Biological Background: CDK6 Function and Localization

  • Cyclin-dependent kinase 6 (CDK6), also known as cell division protein kinase 6 or serine/threonine-protein kinase PLSTIRE, is a member of the CMGC family of serine/threonine kinases.
  • Functions as a key regulator of the cell cycle and differentiation; promotes G1/S transition by phosphorylating substrates such as pRB/RB1 and NPM1.
  • Interacts with D-type cyclins during G1 to form active cyclin D-CDK6 complexes that drive progression through the restriction point and entry into S phase.
  • Plays a dual role in differentiation: generally prevents proliferation and negatively regulates differentiation, yet is essential for the proliferation of specific cell types, including erythroid and hematopoietic cells.
  • Required for adult neurogenesis in the dentate gyrus of the hippocampus and the subventricular zone, likely by modulating G1 length and promoting newborn neuron production.

  • Subcellularly localized to the cytoplasm, nucleus, cell ruffles, cytoskeleton, and centrosomes, consistent with its diverse cellular functions.

  • In Western blotting, expect a band at approximately 37 kDa; optimize dilution and use appropriate controls for confirming band specificity.
  • For immunohistochemistry, consider using heat-induced epitope retrieval and validate staining in known positive tissues (e.g., proliferating areas) and negative controls.
  • If performing immunoprecipitation, use a suitable lysis buffer that preserves protein interactions and verify capture with a control antibody.
