Biological Background: CDK5RAP2 Function and Localization
- CDK5RAP2 (Cep215/C48/MCPH3) is a ~215 kDa centrosomal protein encoded by the CDK5RAP2 gene, which undergoes alternative splicing.
- Acts as a potential regulator of CDK5 activity via interaction with CDK5R1. Related references: PMID:15164053
- Functions as a negative regulator of centriole disengagement, maintaining centriole engagement and cohesion, and is involved in regulating mitotic spindle orientation.
- Plays a role in the spindle checkpoint activation by acting as a transcriptional regulator of BUBR1 and MAD2. Related references: PMID:19282672
- Together with EB1/MAPRE1, promotes microtubule polymerization, bundle formation, growth and dynamics at the plus ends. Related references: PMID:18042621 PMID:17959831 PMID:19553473
- Regulates centrosomal maturation by recruiting the gamma-tubulin ring complex (gTuRC) onto centrosomes. Related references: PMID:18042621 PMID:17959831 PMID:26485573
- Contributes to microtubule nucleation and extension from the centrosome to the cell periphery in complex with PDE4DIP, MAPRE1, and AKAP9, and is required for AKAP9 recruitment to centrosomes. Related references: PMID:29162697
- Localizes to the cytoplasm, centrosome, microtubule organizing center, and Golgi apparatus; widely expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas; mutations are linked to primary microcephaly type 3 (MCPH3).
Experimental Guidance and Technical Tips
- For Western blotting, consider using reducing and denaturing sample preparation due to the large molecular weight (~215 kDa) of CDK5RAP2; include appropriate positive and negative control lysates from human cell lines known to express the target.
- For ELISA, the recombinant immunogen fragment (aa 1744-1893) may serve as a coating standard; validate antibody specificity against full-length protein and relevant controls.
- Given the role of CDK5RAP2 in centrosome biology, immunofluorescence may be a complementary application; optimize fixation and permeabilization conditions for centrosomal antigens.
