Biological Background: CDK5RAP1 Function and Localization

  • CDK5RAP1 (also known as C42, CGI-05, HSPC167, C20orf34) is a mitochondrial methylthiotransferase that modifies four mitochondrial tRNAs (Ser(UCN), Phe, Tyr, Trp) by converting N6-(dimethylallyl)adenosine (i6A) to 2-methylthio-N6-(dimethylallyl)adenosine (ms2i6A) at position 37, which is crucial for translation accuracy. Related references: PMID:22422838 PMID:25738458 PMID:28981754

  • The enzyme specifically inhibits CDK5 activation by its regulator CDK5R1 (p35), implicating a role in neuronal signaling pathways. Related references: PMID:11882646

  • CDK5RAP1 belongs to the radical S-adenosyl-L-methionine (SAM) superfamily and contains a conserved 4Fe-4S cluster essential for its methylthiotransferase activity.

  • Subcellular localization is predominantly mitochondrial, consistent with its function in mitochondrial tRNA modification and protein synthesis.

  • Tissue expression is widespread, with highest levels in brain, placenta, and testis; notably, it is expressed in neurons of the central nervous system.

  • Essential for mitochondrial protein synthesis and respiratory chain function; pathological mutations are associated with mitochondrial disease. Related references: PMID:25738458

  • The protein exists in multiple alternatively spliced isoforms and contains a mitochondrial transit peptide for import.

Experimental Guidance and Technical Tips

  • For Western blot (WB), the predicted molecular weight is approximately 68 kDa; use reducing conditions and optimize loading and antibody dilution for your specific sample.
  • In immunohistochemistry (IHC-P), antigen retrieval may be required; consider testing a range of antibody dilutions to minimize background.
  • For ELISA, the antibody can be employed in direct, indirect, or sandwich formats; pair with a suitable detection system.
  • Cross-reactivity with mouse and rat allows application in rodent models; validate detection specificity in relevant tissue or cell lysates.
