CDK5RAP1 (also known as C42, CGI-05, HSPC167, C20orf34) is a mitochondrial methylthiotransferase that modifies four mitochondrial tRNAs (Ser(UCN), Phe, Tyr, Trp) by converting N6-(dimethylallyl)adenosine (i6A) to 2-methylthio-N6-(dimethylallyl)adenosine (ms2i6A) at position 37, which is crucial for translation accuracy.
