Biological Background: CDK2 Function and Localization

  • CDK2 is a serine/threonine-protein kinase involved in the control of the cell cycle; it is essential for meiosis but dispensable for mitosis, and it plays a critical role in the G1-S transition and G2/M progression. Related references: PMID:10499802, PMID:10884347, PMID:10995386
  • Activated by cyclin E during G1-S and cyclin A during S/G2, CDK2 phosphorylates numerous substrates including RB1, MYC, and NPM1, thereby promoting DNA replication, centrosome duplication, and transcription. Related references: PMID:10499802, PMID:11051553, PMID:19966300
  • CDK2-mediated phosphorylation of BRCA2 controls homologous recombination repair, with low phosphorylation during S phase and increased phosphorylation as cells approach mitosis. Related references: PMID:15800615, PMID:20195506, PMID:21319273
  • It phosphorylates EZH2, promoting H3K27me3 maintenance and epigenetic gene silencing. Related references: PMID:20935635
  • Subcellularly, CDK2 localizes to the cytoplasm, centrosomes, nucleus, Cajal bodies, and endosomes, consistent with its multifaceted roles in cell division and DNA repair.
  • Post-translational modifications such as phosphorylation by the CAK complex, acetylation, and S-nitrosylation dynamically regulate CDK2 activity.

Experimental Guidance and Technical Tips

  • The immunogen was designed from a sequence within amino acids 199-298 of human CDK2. For Western blotting, consider using whole-cell lysates from proliferating cell lines; the predicted bands are around 30-34 kDa.
  • As a KO-validated antibody, it may be suitable for confirming CDK2 knockout efficiency in relevant cell models.
  • For ELISA, consider using recombinant CDK2 protein or cell lysates, and validate against appropriate positive and negative controls.
