Biological Background: CDK19 Function and Localization

  • CDK19, also known as cyclin-dependent kinase 11, CDC2-related protein kinase 6, or cell division cycle 2-like protein kinase 6, is a serine/threonine-protein kinase encoded by the CDK19 gene.
  • It is an integral component of the Mediator complex, serving as a transcriptional coactivator that bridges gene-specific transcription factors with RNA polymerase II to regulate gene expression. CDK19 phosphorylates key substrates including RNA polymerase II and the Notch1 intracellular domain (ICN1), and collaborates with CDK8 in macrophage differentiation. • It localizes to both the cytoplasm and nucleus, with enrichment in the perinuclear region.
  • The protein undergoes post-translational modifications including acetylation and phosphorylation, and is subject to alternative splicing.
  • Mutations in CDK19 are associated with developmental and epileptic encephalopathy 87 (DEE87) and early infantile epileptic encephalopathy 87 (EIEE87), underscoring its neurodevelopmental importance.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 250–350 of human CDK19. This region may be conserved across species, supporting observed cross-reactivity with mouse and rat.
  • For Western blot, a band around 57 kDa is expected; consider using appropriate controls and validate in the relevant cell or tissue lysate.
  • In immunohistochemistry (IHC-P), antigen retrieval conditions may need optimization, and expression patterns should be validated against known cytoplasmic and nuclear localization.
  • For ELISA, use the antibody as a detection reagent in sandwich assays; pairing with a capture antibody may require empirical testing.
