Biological Background: CDK16 Function and Localization
- Cyclin-dependent kinase 16 (CDK16), also known as PCTAIRE1, PCTK1, or serine/threonine-protein kinase PCTAIRE-1, is a member of the CDK family encoded by the CDK16 gene.
- CDK16 plays a role in vesicle-mediated transport and exocytosis; it regulates GH1 release by brain neurons through phosphorylation of NSF, thereby modulating NSF oligomerization (By similarity).
- The kinase is required for normal spermatogenesis and regulates neuron differentiation and dendrite development (By similarity).
- CDK16 is involved in insulin secretion in response to blood glucose changes and can phosphorylate cyclin-Y (CCNY) at Ser-336 in vitro.
- Subcellularly, CDK16 localizes to the cytoplasm, cytoplasmic vesicles, secretory vesicles, cell membrane, and synaptic compartments (synapse/synaptosome).
- At the protein level, CDK16 is detected in pancreas islets, with additional expression in brain and pancreas tissues.
- Keywords for CDK16 include kinase, transferase, phosphoprotein, alternative splicing, and reference proteome, underscoring its regulatory functions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 277–496) of human CDK16, which may target conserved epitopes; consider epitope conservation if using other species.
- For IF/ICC, optimize fixation and permeabilization protocols, as CDK16 associates with multiple membrane compartments.
- In ELISA, validate linearity and specificity using the recombinant immunogen or cell lysates from CDK16-expressing tissues (brain, pancreas).
- CDK16 is a low-abundance kinase; signal amplification or enrichment strategies may be beneficial for detection.
