Biological Background: CDK11B Function and Localization

  • CDK11B, also known as PITSLRE serine/threonine-protein kinase CDC2L1 or p58 CLK-1, is a cyclin-dependent kinase encoded by the CDK11B gene. It exists as multiple isoforms due to alternative initiation and splicing, with some isoforms expressed in a non-cell cycle-dependent manner and others specifically during G2-M phases. Related references: PMID:12082095, PMID:2217177
  • CDK11B acts as a key regulator of pre-mRNA splicing by mediating phosphorylation of SF3B1, which facilitates the association of SF3B1 with U5 and U6 snRNAs in the activated spliceosome, thereby promoting spliceosome assembly. Related references: PMID:36104565, PMID:38059508
  • The kinase also functions as a transcription regulator by phosphorylating serine-2 of the C-terminal domain (CTD) of RNA polymerase II large subunit POLR2A, a critical event for transcriptional elongation and processing. Related references: PMID:32367068, PMID:40858114
  • CDK11B is involved in replication-dependent transcription of histone genes. It binds to histone genes and phosphorylates POLR2A at serine-2 of the CTD, specifically controlling transcriptional elongation of histones and recruitment of 3'-end processing factors. Related references: PMID:32367068
  • As part of a transcription checkpoint upstream of CDK9, CDK11B regulates promoter-proximal pausing of RNA polymerase II, a pause that occurs post-initiation but prior to elongation, likely through CTD phosphorylation. Related references: PMID:40858114
  • During mitosis, CDK11B promotes centromeric transcription to maintain centromeric cohesion, with its kinase activity essential for proper sister chromatid cohesion and chromosome segregation. Related references: PMID:24436071, PMID:38019613
  • Subcellular localization includes the nucleus, chromosomes (including centromeres), and cytoplasm, consistent with its roles in transcription, splicing, and mitosis. The protein is ubiquitously expressed, with some evidence of isoform-specific tissue distribution.
  • Post-translational modifications include phosphorylation (it is activated by the CDK-activating kinase complex comprising CDK7, cyclin-H, and MAT1) and ubiquitination (isopeptide bond and Ubl conjugation), as annotated by the UniProt keyword list. Related references: PMID:41100585

Experimental Guidance and Technical Tips

  • The immunogen was designed as a recombinant fusion protein spanning amino acids 1-130 of human CDK11B (NP_277021.2), targeting the N-terminal region. For IF/ICC, consider testing multiple fixation and permeabilization methods, as CDK11B localizes to the nucleus, chromosomes, and cytoplasm.
  • In ELISA, validate the antibody’s reactivity with the immunogen or relevant cell/tissue lysates under standard assay conditions. Since the antibody is rabbit polyclonal, there may be batch-to-batch variation; it is advisable to titrate the optimal dilution for your specific experimental setup.
  • Cross-reactivity with human, mouse, and rat is expected, but endogenous protein expression levels should be confirmed in your target sample system.
