Biological Background: CDC42 Function and Localization
- CDC42 is a plasma membrane-associated small GTPase that cycles between an active GTP-bound state and an inactive GDP-bound state, regulating various cellular responses.
- It plays a critical role in epithelial cell polarization, bipolar spindle microtubule attachment to kinetochores, and cell migration.

PMID:22843693 - In neurons, CDC42 is involved in the extension and maintenance of filopodia and is required for DOCK10-mediated spine formation in Purkinje cells and hippocampal neurons.
Related references:
PMID:14978216 - It participates in phagocytosis by organizing the F-actin cytoskeleton around phagocytic cups and mediates actin cytoskeletal remodeling during epithelial cell-cell junction formation upon activation by PLEKHG4B.
Related references:
PMID:26465210
PMID:33310911 - The protein localizes to the cell membrane, cytoplasm, centrosomes, spindles, midbody, dendritic projections, and secretory vesicles such as acrosomes.
- Post-translational modifications include phosphorylation, prenylation, methylation, and ubiquitination, which regulate its subcellular targeting and activity.
- Alternative splicing generates multiple isoforms, and mutations in CDC42 are associated with intellectual disability.
- The predicted molecular weight is approximately 21.3 kDa (observed at ~21 kDa in SDS-PAGE).
Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was raised against a recombinant protein corresponding to amino acids 1-186 of human CDC42 (isoform 1). It is expected to recognize endogenous CDC42 in human samples.
- For immunofluorescence/immunocytochemistry (IF/ICC), a typical starting dilution might be considered; however, optimal working concentration should be empirically determined using appropriate positive and negative controls.
- For ELISA, the antibody can be used for antigen capture or detection; pairing with a matched detection antibody may improve sensitivity.
- Given the antibody's polyclonal nature, lot-to-lot variability may occur; validation in the relevant biological system is advisable.
- The predicted band size in Western blot is ~21 kDa, although this application is not listed; cross-reactivity with other Rho family GTPases cannot be excluded.
