Biological Background: CDC20 Function and Localization

  • CDC20 (cell division cycle protein 20 homolog), also known as p55CDC, is a substrate-specific adapter and activator of the anaphase promoting complex/cyclosome (APC/C) that plays an essential role in the metaphase/anaphase transition of the cell cycle. As part of the APC/C, it targets key substrates such as cyclin B and securin for ubiquitination and degradation, thereby driving anaphase onset and mitotic exit. Related references: PMID:9734353 PMID:27030811 PMID:29343641 PMID:27120157 PMID:27509861 PMID:32666501

  • CDC20 recognizes and binds specific degradation motifs (D box, KEN box, and ABBA motifs) on protein substrates, conferring substrate specificity to the APC/C complex. Related references: PMID:29343641 PMID:27509861

  • CDC20 is a key component of the mitotic checkpoint complex (MCC), which includes MAD2L1/MAD2, BUB1B/BUBR1, and BUB3. The MCC inhibits APC/C activity until all chromosomes are properly attached to the mitotic spindle, ensuring accurate chromosome segregation. Related references: PMID:27509861 PMID:9811605 PMID:9637688

  • Beyond its canonical mitotic functions, CDC20-APC/C has been implicated in postmitotic neuronal processes, including positive regulation of synaptic vesicle clustering and presynaptic differentiation, as well as promotion of proper radial migration through degradation of CCDC41.

  • Subcellularly, CDC20 localizes to the cytoplasm, centrosome, kinetochore, and spindle pole, reflecting its dynamic roles during mitosis and checkpoint signaling.

  • CDC20 is post-translationally regulated by phosphorylation, acetylation, and ubiquitination, and contains WD repeat domains characteristic of its protein family. These modifications modulate its activity and stability throughout the cell cycle.

Experimental Guidance and Technical Tips

  • The antibody was raised against a recombinant fusion protein corresponding to the N-terminal region (amino acids 1-100) of human CDC20. For Western blot, the predicted molecular weight is approximately 55 kDa.
  • For immunohistochemistry (IHC-P), consider optimizing antigen retrieval conditions; test with a tissue known to have high mitotic index (e.g., tonsil or tumor) as a positive control.
  • For immunofluorescence (IF/ICC), fixation and permeabilization protocols may affect staining patterns; validate with appropriate controls.
  • This polyclonal antibody may exhibit lot-to-lot variability; inclusion of a knockout or knockdown control is recommended to confirm specificity in your experimental system.
