Biological Background: CDC16 Function and Localization
- CDC16 (APC6) is a core subunit of the anaphase promoting complex/cyclosome (APC/C), a multi-subunit E3 ubiquitin ligase essential for cell cycle progression. Related references: PMID:18485873
- APC/C mediates ubiquitination and subsequent proteasomal degradation of key cell cycle regulators, primarily catalyzing 'Lys-11'-linked polyubiquitin chains, and to a lesser extent 'Lys-48'- and 'Lys-63'-linked chains. It can also assemble branched 'Lys-11'/'Lys-48' chains on substrates. Related references: PMID:18485873, PMID:29033132
- Activation of APC/C by CDC20 in metaphase triggers the degradation of securin and cyclin B, promoting the transition to anaphase and mitotic exit. Related references: PMID:27120157, PMID:27509861
- APC/C activity is inhibited by the mitotic checkpoint complex (MCC) to ensure accurate chromosome segregation until all kinetochores are properly attached. Related references: PMID:27509861
- CDC16 localizes to the cytoplasm, centrosomes, and mitotic spindle during cell division, consistent with its role in mitotic regulation.
- CDC16 belongs to a family of APC/C subunits characterized by tetratricopeptide repeat (TPR) domains that mediate protein-protein interactions.
- The protein undergoes phosphorylation, which may regulate its activity or interactions within the complex.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 200-300 of human CDC16. This internal sequence may be suitable for detecting full-length CDC16 in western blot applications, though not included in the standard application list, consider validating for denaturing protein analysis given the predicted molecular weight of ~72 kDa.
- For IHC-P, consider using heat-induced antigen retrieval (e.g., citrate buffer, pH 6.0) and optimise primary antibody concentration.
- In IF/ICC, ensure proper fixation (e.g., 4% paraformaldehyde) and permeabilization to allow access to cytoplasmic and centrosomal CDC16.
- ELISA may be used to quantify CDC16; titrate the antibody and validate cross-reactivity with your target species (human, mouse, rat).
