Biological Background: CDC123 Function and Localization
- CDC123, also known as cell division cycle protein 123 homolog, protein D123, HT-1080, or PZ32, is a translation initiation factor eIF2 assembly protein.
- It acts as an ATP-dependent protein-folding chaperone for the eIF2 complex, a crucial step in translation initiation. Related references: PMID:35031321, PMID:37507029
- By binding to the gamma subunit, CDC123 enables the eIF2 complex to assemble with the alpha and beta subunits.
- The protein is localized in the cytoplasm.
- CDC123 is widely expressed, with the highest expression observed in testis; it is also present in spleen, thymus, prostate, ovary, small intestine, colon, and leukocytes.
- Key features include ATP-binding, chaperone activity, and metal ion binding (magnesium); it is also identified as a phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering the full-length human CDC123 sequence (amino acids 1–336). For western blotting, a theoretical band around 39 kDa may be observed; consider using a 10% SDS-PAGE gel for optimal resolution.
- For ELISA applications, test the antibody as a capture or detection reagent after appropriate pairing and optimization in your assay system.
- CDC123 is a widely expressed cytoplasmic chaperone. If the target protein abundance is low, sample enrichment or increased loading may be necessary.
- Cross-reactivity is reported for human, mouse, and rat; validate in your specific sample lysate to confirm detection.
