Biological Background: CDA Function and Localization
- CDA (Cytidine deaminase), also known as CDD or cytidine aminohydrolase, is a zinc-dependent hydrolase that scavenges exogenous and endogenous cytidine and 2'-deoxycytidine, converting them to uridine and deoxyuridine for UMP synthesis.
- It plays a critical role in nucleotide metabolism and the pyrimidine salvage pathway.
- Highly expressed in granulocytes, with very low expression in fibroblasts, chondrocytes, monocytes, and T- and B-cell lines, suggesting a specialized function in myeloid cells.
- The enzyme requires zinc as a metal cofactor for its catalytic activity.
- Belongs to the cytidine and deoxycytidylate deaminase family; 3D structure has been determined.
- Predicted molecular weight of ~16 kDa.
Experimental Guidance and Technical Tips
- The immunogen used is full-length human CDA recombinant protein, so the antibody is expected to recognize the native protein across applications.
- For Western blotting, a band at ~16 kDa corresponding to CDA monomer may be observed; however, note that endogenous expression is tissue-specific, with high levels in granulocytes and low levels in other cell types.
- In IHC-P, consider using granulocyte-rich tissues (e.g., bone marrow, spleen) as positive controls, and validate staining with appropriate negative controls.
- The antibody cross-reacts with mouse CDA; for experiments using mouse samples, validate in the relevant mouse tissue/cell type, considering potential differences in expression patterns.
- ELISA applications may require optimization of coating antigen and antibody dilutions.
