Biological Background: CD98/SLC3A2 Function and Localization

  • CD98/SLC3A2 (4F2 heavy chain) is a type II transmembrane glycoprotein that acts as a chaperone for SLC7 family light chains, forming functional heterodimeric amino acid transporters at the plasma membrane. Related references: PMID:10574970, PMID:10903140, PMID:11557028

  • For Western blotting, use reducing conditions; the core protein is ~58 kDa, but glycosylation can increase the apparent molecular weight. Validate using human tissue or cell line lysates.
  • In immunofluorescence/immunocytochemistry, expect membrane and intracellular vesicular (lysosome) staining patterns. Permeabilization may be required for detection of intracellular epitopes.
  • For ELISA, the antibody can be tested as a capture or detection reagent; pair with a suitable detection antibody for sandwich ELISA development.
  • Cross-reactivity with human CD98 has been confirmed; test for cross-reactivity in other species if required for your assay.
