Biological Background: CD98/SLC3A2 Function and Localization

• CD98/SLC3A2 (4F2hc) is a type II transmembrane glycoprotein encoded by the SLC3A2 gene. It undergoes extensive post-translational modifications including N-glycosylation, disulfide bond formation, acetylation, phosphorylation, and ubiquitination.
• SLC3A2 acts as a chaperone, forming heterodimers with SLC7 family amino acid transporters (SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A10, SLC7A11) and is essential for their plasma membrane localization, stability, and transport activity. • The immunogen covers amino acids 206-630 of human SLC3A2 (NP_002385.3), which includes the majority of the extracellular domain. This region may be used for epitope mapping or competition experiments.
• For Western blotting, a band between 58-71 kDa is expected due to glycosylation. Validate with positive control lysates from human, mouse, or rat cell lines; consider using deglycosylation enzymes to confirm specificity.
• In immunofluorescence and immunohistochemistry, test the antibody at several dilutions and include appropriate controls (e.g., isotype control, secondary-only) to minimize background. Note that SLC3A2 localizes to the plasma membrane and intracellular compartments.
• The antibody may cross-react with mouse and rat SLC3A2 based on sequence homology; verify in the relevant sample system before use.
