Biological Background: CD98 Function and Localization
-
CD98, also known as SLC3A2 or 4F2hc, is a type II transmembrane glycoprotein that serves as the heavy chain of several amino acid transporters, essential for their biogenesis and plasma membrane localization.
-
It forms heterodimers with multiple SLC7 family light chains (e.g., SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A10, SLC7A11), thereby determining substrate specificity for the transport of neutral, dibasic, and anionic amino acids, as well as thyroid hormones and L-DOPA.

-
CD98 localizes primarily to the plasma membrane, with enrichment at apical and basolateral domains in polarized epithelia, and is also found at cell junctions, on lysosomal and melanosomal membranes.
-
Beyond transport, CD98 modulates integrin-dependent signaling, promoting cell adhesion, spreading, migration, and tumor progression.
-
Post-translational modifications include disulfide bond formation, N-linked glycosylation, phosphorylation, and isopeptide linkage, which may regulate its stability and interactions.
-
CD98 is widely expressed in adult tissues, with highest levels in kidney, small intestine, spleen, thymus, and liver; in the brain, it is detected on capillary endothelial cells.

Experimental Guidance and Technical Tips
- The immunogen spans amino acids 105–526 of mouse CD98, covering a large extracellular and transmembrane region; this design may yield recognition of multiple epitopes and isoforms.
- Validated applications include Western blotting and ELISA; for WB, consider standard denaturing and reducing conditions to detect the ~58 kDa band, and validate in your sample type.
- Cross-reactivity with rat CD98 is confirmed, making this antibody suitable for studies in both mouse and rat models; always include appropriate controls.
