Biological Background: CD97 Function and Localization
- CD97 (also known as Adhesion G protein-coupled receptor E5 or leukocyte antigen CD97) is encoded by the Adgre5 gene in mouse.
- It functions as a receptor involved in both adhesion and signaling processes, playing an essential role in leukocyte migration.
- CD97 belongs to the adhesion G protein-coupled receptor (GPCR) family, characterized by a large extracellular domain with EGF-like repeats that facilitate cell-cell interactions.
- At the protein level, CD97 is subject to alternative splicing, glycosylation, and phosphorylation, and it contains calcium-binding EGF-like domains and disulfide bonds critical for its structural integrity.
- Subcellularly, CD97 localizes to the cell membrane and can also be secreted into the extracellular space.
- Although predominantly expressed by immune cells, CD97 is ubiquitously found with particularly high expression in the lung and thymus gland; in spleen, it is detected on most myeloid cells and variable portions of T-cells, B-cells, and NK cells, while in bone marrow it is present in nearly all myeloid cells but largely absent from erythroid cells.

- For western blotting, an expected band around 90 kDa may be observed; due to glycosylation and potential disulfide-linked structures, consider running samples under both reducing and non-reducing conditions to resolve the target.
- For ELISA applications, the antibody can be used as a capture or detection reagent; optimize coating concentration and detect with appropriate secondary antibodies.
