Biological Background: CD97 (Adhesion G protein-coupled receptor E5) Function and Localization

  • CD97, also known as Adhesion G protein-coupled receptor E5 or ADGRE5, is a member of the adhesion GPCR family characterized by a large extracellular domain with EGF-like repeats.
  • The receptor is potentially involved in both adhesion and signaling processes early after leukocyte activation, playing an essential role in leukocyte migration.
  • CD97 is broadly expressed on most hematopoietic cells, including activated lymphocytes, monocytes, macrophages, dendritic cells, and granulocytes, as well as smooth muscle cells and various carcinomas.
  • Subcellularly, CD97 localizes to the cell membrane and can also be found as a secreted form in the extracellular space.
  • Expression is increased under inflammatory conditions, such as in the CNS of multiple sclerosis and in synovial tissue of rheumatoid arthritis, with detectable soluble CD97 in synovial fluid.
