Biological Background: CD97 Function and Localization
- CD97, also known as Adhesion G protein-coupled receptor E5 or Leukocyte antigen CD97, is encoded by the ADGRE5 gene.
- Functions as an adhesion G protein-coupled receptor involved in both adhesion and signaling processes early after leukocyte activation, playing an essential role in leukocyte migration.
- Contains EGF-like domains, a GPCR proteolytic site, and a transmembrane helix characteristic of the adhesion GPCR family.
- Localized predominantly at the cell membrane and is also found secreted in the extracellular space.
- Broadly expressed on most hematopoietic cells, including activated lymphocytes, monocytes, macrophages, dendritic cells, and granulocytes; also expressed by smooth muscle cells and various carcinomas.
- Expression is upregulated under inflammatory conditions, such as in the CNS of multiple sclerosis and synovial tissue of rheumatoid arthritis, with detectable soluble CD97 in synovial fluid.
- Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation.
