Biological Background: CD93 Function and Localization

  • CD93 is a type I transmembrane glycoprotein also known as complement component C1q receptor, lymphocyte antigen 68 (Ly-68), AA4, C1qRp, and C1qr1. It belongs to the group of defense collagen receptors and contains EGF-like domains, glycosylation sites, and disulfide bonds.
  • Acts as a cell surface receptor involved in various physiological processes, including inflammation, phagocytosis, and cell adhesion, and plays a key role in the immune response. Related references: PMID:15004139 PMID:20439917 PMID:38441970
  • Functions as a phagocytic receptor by binding defense collagens such as C1q, mannose-binding lectin (MBL2), and surfactant protein A (SFTPA1), thereby enhancing the clearance of apoptotic cells and immune complexes. Related references: PMID:15004139
  • Plays a critical role in angiogenesis and endothelial cell function by associating with beta-dystroglycan, leading to SRC-dependent phosphorylation and recruitment of CBL, which then provides a docking site for downstream signaling components like CRKL to promote cell migration. It also serves as a receptor for extracellular matrix ligands multimerin-2 (MMRN2) and IGFBP7.
  • Regulates endothelial barrier integrity by modulating VEGFR2 signaling, thereby controlling vascular permeability. Related references: PMID:38441970
  • Predominantly localized to the cell membrane; expressed in lung, heart, bone marrow, and macrophages, with lower levels in ovary and embryo, but absent in brain, adult liver, thymus, and epithelial cells.
  • The predicted molecular weight is approximately 69 kDa, consistent with the observed 69 kDa band in SDS-PAGE.

Experimental Guidance and Technical Tips

  • The immunogen encompasses amino acids 23–181 of mouse CD93, which corresponds to the N-terminal extracellular domain containing the C1q binding region and EGF-like repeats; this region is expected to be exposed on the cell surface and may also be present in soluble forms.
  • For flow cytometry (FC), consider using this antibody to detect cell surface CD93, as the target is a transmembrane receptor. Validation in relevant cell lines or primary macrophages is recommended.
  • In immunofluorescence/immunocytochemistry (IF/ICC), membrane staining should be observed; ensure appropriate fixation and permeabilization conditions to preserve epitope integrity.
  • For ELISA applications, the recombinant immunogen may serve as a capture standard, and the antibody can be used for sandwich assays if paired with a suitable detection antibody. Sensitivity to the native protein should be confirmed.
  • Cross-reactivity with human CD93 indicates potential utility in human cell systems; however, affinity and specificity should be verified in each experimental context.
