Biological Background: CD90/Thy1 Function and Localization
- CD90 (Thy-1) is a cell surface glycoprotein also designated CDw90, encoded by the THY1 gene in humans (UniProt P04216).
- It belongs to the immunoglobulin superfamily, characterized by an immunoglobulin-like domain, and is anchored to the plasma membrane via a glycosylphosphatidylinositol (GPI) anchor.
- The protein undergoes several post-translational modifications, including N-linked glycosylation, disulfide bond formation, phosphorylation, and the addition of a GPI anchor; the signal peptide is cleaved and the N-terminal glutamine cyclized to pyroglutamic acid.
- Subcellularly, CD90 is localized to the cell membrane where it may participate in cell-cell and cell-ligand interactions.
- Functional studies suggest a role in synaptogenesis and other events in the brain, potentially mediating adhesion and signaling between neurons and glial cells.
- The mature protein has a predicted molecular weight of approximately 18 kDa.

- The immunogen used is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human CD90. When planning experiments, consider that the antibody may recognize epitopes within this region.
- For western blot (WB), the reported molecular weight is ~18 kDa. Use appropriate gel percentages and transfer conditions for small proteins. Include positive controls such as human or rat brain lysates.
- In immunohistochemistry (IHC-P), antigen retrieval conditions may require optimization; test both heat-induced and enzymatic methods on formalin-fixed paraffin-embedded sections.
