Biological Background: CD9 Function and Localization
- CD9 (also known as MIC3, MRP-1, TSPAN29) is a member of the tetraspanin superfamily, a group of four-transmembrane domain proteins that organize membrane microdomains and modulate cell adhesion, migration, and signaling.
- CD9 is an integral membrane protein that associates with integrins and other tetraspanins to regulate diverse processes, including sperm-egg fusion, platelet activation and aggregation, and cell adhesion (PubMed:14575715, PubMed:18541721, PubMed:8478605). Related references: PMID:14575715, PMID:18541721, PMID:8478605
- In macrophages, CD9 associates with CD81 and beta-1/beta-2 integrins to prevent macrophage fusion into multinucleated giant cells specialized for ingesting complement-opsonized large particles (PubMed:12796480). Related references: PMID:12796480
- CD9 is involved in inhibiting myotube fusion during muscle regeneration (by similarity) and prevents the fusion of mononuclear cell progenitors into osteoclasts responsible for bone resorption (by similarity).
- CD9 acts as a receptor for PSG17 (by similarity) and regulates paranodal junction formation (by similarity).
- The CD9 protein contains disulfide bonds, is palmitoylated on juxtamembrane cysteine residues, and is annotated as a glycoprotein—features that contribute to its membrane organization and intermolecular interactions.
- Subcellularly, CD9 localizes to the cell membrane, is secreted in extracellular exosomes, and is detected in platelets as well as various hematopoietic and epithelial cells (PubMed:19640571). Related references: PMID:19640571
- CD9 plays a role in tumor metastasis and cell motility, with implications in cancer biology (PubMed:7511626, PubMed:8478605). Related references: PMID:7511626, PMID:8478605
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human CD9 protein (amino acids 1–228), which may allow the antibody to recognize both extracellular and intracellular epitopes; perform epitope mapping if domain-specific binding is required.
- For Western blot, a band near 25 kDa is expected, though post‑translational modifications such as glycosylation and palmitoylation may cause a molecular weight shift; validate with appropriate positive and negative controls.
- In immunofluorescence and immunohistochemistry, CD9 is expressed on the plasma membrane and in intracellular vesicles; include permeabilization steps for intracellular staining, and consider co‑staining with organelle markers to confirm localization.
- For flow cytometry, use human cell lines or primary cells known to express CD9 (e.g., platelets, hematopoietic cells) as positive controls; Fc receptor blocking can reduce non‑specific binding.
- This antibody is suitable for ELISA; for sandwich ELISA, consider pairing it with a detection antibody that targets a non‑overlapping epitope.
