  • CD8B (CD8beta) is the beta chain of the CD8 coreceptor, an immunoglobulin domain-containing transmembrane glycoprotein essential for adaptive immunity.
  • It functions as a coreceptor for MHC class I molecules, interacting simultaneously with the T-cell receptor (TCR) and the peptide-MHC complex on antigen-presenting cells.
  • Upon engagement, the cytoplasmic tail recruits the Src kinase LCK to the TCR-CD3 complex, initiating downstream signaling cascades that promote cytokine production, adhesion, and cytotoxic T-lymphocyte activation.
  • Palmitoylation of the cytoplasmic tail targets CD8B to lipid rafts, enriching signaling proteins and enhancing activation efficiency.
  • CD8B is critically required for thymic selection and maturation of CD8+ T-cells.
  • Multiple isoforms (1, 3, 5, 6, 7, 8) are generated by alternative splicing and expressed in both thymic and peripheral CD8+ T-cells, with isoform 1 predominant in thymus and isoform 6 in periphery.
