Biological Background: CD8A Function and Localization
- CD8A encodes the alpha chain of the T-cell surface glycoprotein CD8, which belongs to the immunoglobulin superfamily and is alternatively spliced.
- It functions as a coreceptor for MHC class I molecules on antigen-presenting cells, interacting simultaneously with the TCR and MHC class I to facilitate T-cell activation.
- Upon antigen recognition, CD8A recruits the Src kinase LCK to the TCR-CD3 complex, initiating downstream signaling pathways that lead to lymphokine production, motility, adhesion, and cytotoxic T-lymphocyte (CTL) activation.
- CD8A can form heterodimers with CD8B on thymus-derived T-cells or homodimers on subsets of natural killer cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
- In NK cells, CD8A homodimers at the cell surface promote survival and enable conjugation and lysis of multiple target cells.
- CD8A is localized primarily on the cell membrane but can also be secreted.
- Expressed in plasmacytoid dendritic cells upon herpes simplex virus-1 stimulation, enhancing immune surveillance.
- The protein is modified by disulfide bonds, glycosylation, and palmitoylation, reflecting its integral membrane nature.
