Biological Background: CD8A Function and Localization

  • CD8A encodes the alpha chain of the T-cell surface glycoprotein CD8, which belongs to the immunoglobulin superfamily and is alternatively spliced.
  • It functions as a coreceptor for MHC class I molecules on antigen-presenting cells, interacting simultaneously with the TCR and MHC class I to facilitate T-cell activation.
  • Upon antigen recognition, CD8A recruits the Src kinase LCK to the TCR-CD3 complex, initiating downstream signaling pathways that lead to lymphokine production, motility, adhesion, and cytotoxic T-lymphocyte (CTL) activation.
  • CD8A can form heterodimers with CD8B on thymus-derived T-cells or homodimers on subsets of natural killer cells, memory T-cells, intraepithelial lymphocytes, monocytes, and dendritic cells.
  • In NK cells, CD8A homodimers at the cell surface promote survival and enable conjugation and lysis of multiple target cells.
  • CD8A is localized primarily on the cell membrane but can also be secreted.
  • Expressed in plasmacytoid dendritic cells upon herpes simplex virus-1 stimulation, enhancing immune surveillance.
  • The protein is modified by disulfide bonds, glycosylation, and palmitoylation, reflecting its integral membrane nature.

Experimental Guidance and Technical Tips

  • The immunogen comprising the extracellular domain (aa 22-182) suggests the antibody likely recognizes the native conformation of CD8A, making it suitable for applications that require detecting cell surface proteins, such as IF-P and IHC-P.
  • For Western blotting, consider the heterodimeric nature of CD8, as multiple bands around 21-30 kDa may be observed due to glycosylation or dimerization; validate in relevant cell lysates.
  • When used in multiplex immunohistochemistry (mIHC), consider pairing with other immune cell markers to profile T-cell subsets in tissue sections.
  • Due to cross-reactivity among human, mouse, rat, and cynomolgus monkey, this antibody can be utilized for translational studies; however, always verify species-specific reactivity in your experimental system.
