Biological Background: CD8A Function and Localization
- CD8A is the alpha chain of the CD8 coreceptor, also known as Lyt-2 or Ly-2, and belongs to the immunoglobulin superfamily.
- It functions as a coreceptor for MHC class I molecules by binding to the α3 domain of MHC I while simultaneously interacting with the T-cell receptor (TCR) on cytotoxic T lymphocytes.
- In T cells, CD8A recruits the Src kinase LCK to the TCR-CD3 complex, initiating phosphorylation cascades that lead to lymphokine production, adhesion, motility, and activation of cytotoxic T-lymphocytes (CTLs).
- In NK cells, CD8A homodimers provide a survival mechanism that enables conjugation and lysis of multiple target cells, and promote differentiation of activated lymphocytes into memory CD8 T cells.
