Biological Background: CD8A Function and Localization

  • CD8A encodes the alpha chain of the T-cell surface glycoprotein CD8, also known as T8/Leu-2, which is a coreceptor for MHC class I molecules. The CD8 protein exists as either a disulfide-linked heterodimer of alpha/CD8A and beta/CD8B chains or less commonly as a CD8A homodimer.
  • CD8A is essential for the immune response. In T-cells, it interacts with the T-cell receptor (TCR) and MHC class I:peptide complexes on antigen-presenting cells, recruiting the Src kinase LCK to initiate signaling pathways that lead to T-cell activation, proliferation, and differentiation into cytotoxic T-lymphocytes (CTLs).
  • CD8A enables CTLs to recognize and eliminate infected and tumor cells. The intracellular signaling cascade triggered by CD8A-LCK phosphorylation promotes lymphokine production, motility, adhesion, and target cell lysis.
  • On natural killer (NK) cells, CD8A homodimers provide a survival mechanism that facilitates conjugation with and lysis of multiple target cells. CD8A homodimers also support the survival and differentiation of activated lymphocytes into memory CD8+ T-cells.
