Biological Background: CD86 Function and Localization
- CD86 (also known as B7-2, B70, and FUN-1) is a T-lymphocyte activation antigen belonging to the immunoglobulin superfamily that functions as a costimulatory molecule essential for T-cell activation. Related references: PMID:12196291 PMID:7694363
- It serves as the primary auxiliary signal augmenting the MHC/TCR signal in naïve T-cells by binding to the CD28 receptor constitutively expressed on T-cells, playing a critical role in the early T-cell activation decision between immunity and anergy. Related references: PMID:12196291 PMID:7694363 PMID:7527824
- CD86 also acts as an inhibitor of T-cell activation by serving as a ligand for CTLA4, a decoy receptor that blocks CD28-mediated priming, thereby providing a negative regulatory checkpoint. Related references: PMID:11279501
- It is involved in B-cell regulation and modulates IgG1 production; upon CD40 engagement, activates NF-κB signaling through phospholipase C and protein kinase C.
- CD86 is a single-pass type I membrane protein localized on the cell surface of antigen-presenting cells, with a molecular weight of ~38 kDa (precursor ~33 kDa).
- It is post-translationally modified by N-linked glycosylation, disulfide bond formation, and ubiquitination, and exists in alternatively spliced isoforms.
- CD86 serves as a receptor for adenovirus subgroup B, facilitating viral entry during infection.
- The protein is classified under adaptive immunity, host-virus interaction, and immunoglobulin domain proteins.
Experimental Guidance and Technical Tips
- For Western blot (WB) analysis, consider using whole-cell lysates from human, mouse, or rat antigen-presenting cells; the antibody may detect a band around 38–50 kDa due to variable glycosylation. Validate the signal with a relevant positive control.
- When performing immunofluorescence (IF/ICC, IF-P), expect membrane localization; use a suitable fixation and permeabilization protocol to preserve the epitope.
- For ELISA applications, this polyclonal antibody can be explored as a capture or detection reagent for recombinant or native CD86; optimization of working concentrations is recommended.
- Given the polyclonal nature, lot-to-lot consistency should be verified for quantitative assays; consider pairing with a monoclonal antibody for critical applications.
