Biological Background: CD86 Function and Localization

  • CD86 (B7-2, LAB72) is a costimulatory molecule belonging to the immunoglobulin superfamily that plays an essential role in T-lymphocyte activation by acting as a ligand for the CD28 receptor, providing the primary auxiliary signal for naive T-cell priming.   • This molecule is critical in the early decision between T-cell immunity and anergy, occurring within 24 hours of activation, thereby influencing adaptive immune responses. Related references: PMID:7527824

  • CD86 also functions as an inhibitor of T-cell activation by binding CTLA4, which outcompetes CD28 and blocks CD28-mediated costimulation, acting as a decoy receptor. Related references: PMID:11279501

  • In B cells, CD86 engagement with CD40 activates the NF-kappa-B signaling pathway via phospholipase C and protein kinase C, regulating IgG1 production (By similarity).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 20–241, corresponding to the extracellular domain of human CD86. This antibody is expected to recognize the native surface protein, making it suitable for flow cytometry (FC) and immunofluorescence/immunocytochemistry (IF/ICC) on live cells.
  • For western blot (WB), CD86 migrates at ~38 kDa. Consider using reducing conditions and validate the band in stimulated antigen-presenting cells (e.g., dendritic cells or B cells) where CD86 is upregulated.
  • In ELISA applications, this antibody may be used for capture or detection of soluble CD86 or in cell-based assays. Titration and pairing with other clones are recommended.
  • The antibody is specific for human CD86. Cross-reactivity with mouse or rat CD86 has not been reported; validate in the relevant sample system if needed.
  • To enhance detection, activation of antigen-presenting cells with stimuli such as LPS, CD40L, or cytokines can significantly upregulate CD86 surface expression.
