Biological Background: CD85j (LILRB1) Function and Localization

  • LILRB1 (CD85j) is a leukocyte immunoglobulin-like receptor subfamily B member that functions as an inhibitory receptor for a broad range of MHC class I molecules, including HLA-A, HLA-B, HLA-C, HLA-G, and HLA-F.
    Related references: PMID:16455647 PMID:28636952

  • It also recognizes the human cytomegalovirus class I MHC homolog H301/UL18, contributing to viral immune evasion.

  • Ligand binding to LILRB1 triggers inhibitory signals that downregulate immune responses, including inhibition of FCER1A-mediated serotonin release and FCGR1A-mediated protein phosphorylation and calcium mobilization.
    Related references: PMID:11907092 PMID:9285411 PMID:9842885

  • Engagement of LILRB1 on natural killer (NK) cells or T cells by MHC class I molecules protects target cells from lysis.

  • In decidual NK cells, LILRB1 interaction with HLA-G/β2-microglobulin/peptide complexes induces secretion of growth-promoting factors, supporting maternal-fetal tolerance.
    Related references: PMID:19304799 PMID:29262349

  • LILRB1 signaling reprograms B cells toward an immune suppressive phenotype.
    Related references: PMID:24453251

  • LILRB1 is expressed on the cell membrane and also exists as a secreted form. It is found on B cells, monocytes, dendritic cells, decidual macrophages, and decidual NK cells.

  • The protein is a transmembrane glycoprotein with immunoglobulin domains, disulfide bonds, and phosphorylation sites, classified under the immunoglobulin superfamily.

Experimental Guidance and Technical Tips

  • For Western blotting, the predicted molecular weight is approximately 71 kDa; however, due to glycosylation, the observed band may migrate at a higher apparent molecular weight. Use reducing conditions and validate with relevant human cell lysates (e.g., B cells or monocytes).
  • For immunofluorescence/immunocytochemistry, fixation and permeabilization protocols should be optimized to detect membrane-localized LILRB1. Consider co-staining with cell surface markers for confirmation.
  • In ELISA applications, this polyclonal antibody can serve as capture or detection reagent. Pairing with a monoclonal antibody may improve specificity in sandwich ELISA setups.
  • The antibody is specific to human LILRB1; cross-reactivity with other species is not expected. Validate in the relevant sample system to confirm performance.
  • As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative assays.
