Biological Background: CD84 Function and Localization
- Protein identity and gene family: CD84 (also known as SLAMF5, LY9B, hCD84, mCD84) is a member of the signaling lymphocytic activation molecule (SLAM) family, which are immunoglobulin superfamily receptors involved in immune regulation. It is encoded by the CD84 gene on human chromosome 1.
- Self-ligand receptor and immune modulation: As a self-ligand receptor, CD84 modulates activation and differentiation of various immune cells through homo- or heterophilic interactions. Its activity is governed by cytoplasmic adapters SH2D1A/SAP and SH2D1B/EAT-2, linking it to both innate and adaptive immunity. It enhances NK cell cytotoxicity, T-cell proliferation, and homophilic interactions boost IFNG secretion and platelet stimulation.
- Hematopoietic progenitor cell marker: CD84 serves as a marker for hematopoietic progenitor cells. Related references: PMID:11564780 PMID:12115647 PMID:12928397
- Germinal center function and B-cell tolerance: CD84 is required for prolonged T-cell:B-cell contact, optimal T follicular helper function, and germinal center formation. In germinal centers, it maintains B-cell tolerance and helps prevent autoimmunity (By similarity).
- Negative regulation in mast cells: In mast cells, CD84 negatively regulates high-affinity immunoglobulin epsilon receptor signaling, independent of SH2D1A and SH2D1B, but involving FES and PTPN6/SHP-1. Related references: PMID:22068234
- Positive regulation of autophagy in dendritic cells: CD84 positively regulates macroautophagy in primary dendritic cells by stabilizing IRF8 and inhibiting its TRIM21-mediated proteasomal degradation. Related references: PMID:29434592
- Subcellular localization and tissue distribution: CD84 is a single-pass type I membrane protein localized to the cell membrane. It is predominantly expressed in hematopoietic tissues, including lymph node, spleen, peripheral leukocytes, macrophages, B-cells, monocytes, platelets, thymocytes, T-cells, and dendritic cells. Memory T-cells show high expression.
- Post-translational modifications and structural features: CD84 is a glycoprotein with a signal peptide, multiple immunoglobulin domains, and conserved disulfide bonds. It is known to be phosphorylated, as reflected in the keywords.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 22-225, corresponding to the extracellular domain of CD84, making this antibody suitable for detecting surface and soluble forms in applications like IF/ICC and ELISA.
- For Western blot, consider using reducing conditions and loading 20–30 µg of cell lysate from human immune cell lines (e.g., Jurkat, THP-1) per lane; the expected band appears around 39 kDa.
- For immunofluorescence, fixation with 4% paraformaldehyde and permeabilization with 0.1% Triton X-100 may be required to detect intracellular pools; validate with appropriate controls.
- For ELISA, pairing with a matched detection antibody may enable development of a sandwich ELISA to quantitate soluble CD84 in serum or culture supernatant; optimize coating concentrations.
- As a polyclonal antibody, this product may exhibit lot-to-lot variability; we recommend validating each lot in your specific assay system.
- CD84 expression can be modulated by immune stimuli; verify target expression levels under your experimental conditions before use.
